چکیده
Abstract Background We previously reported the results of CRISPR/Cas9 knock-out (KO) type-I and type-III vitellogenins (Vtgs) in zebrafish, which provided first experimental evidence on essentiality disparate functioning Vtgs at different stages during early development. However, specific contributions types Vtg to major cellular processes remained be investigated. The present study employed liquid chromatography tandem mass spectrometry (LC-MS/MS) meet this deficit. Proteomic profiles zebrafish eggs lacking three simultaneously ( vtg1 -KO), or only type III vtg3 -KO) were compared those wild (Wt) eggs. Obtained spectra searched against a proteome database identified proteins quantified based normalized spectral counts. Results vtg -KO caused severe changes 1-cell stage These disclosed by molecular signatures that highly resembled proteomic phenotype poor quality our prior studies. perturbations abundances hundreds revealed unique, noncompensable multiple protein energy homeostasis. lack contribution appears have significant impact endoplasmic reticulum mitochondrial functions, thus embryonic development, even after zygotic genome activation. Increased stress, Redox/Detox activities, glycolysis/gluconeogenesis, enrichment proliferation human neurodegenerative disease related activities both - found indicators aforementioned conditions. Distinctive increase apoptosis Parkinson pathways, as well decrease lipid metabolism implies compelling roles Vtg3, least abundant form vertebrate eggs, activities. Several differentially representing altered mechanisms been strong candidate markers for studying details these development possibly other vertebrates. Conclusions findings indicate global egg is subject extensive modification depending presence absence modifications can developmental competence.

Scrambled eggs: Proteomic portraits and novel biomarkers of egg quality in zebrafish (Danio rerio)
Egg quality is a complex biological trait and a major determinant of reproductive fitness in all animals. This study delivered the first proteomic portraits of egg quality in zebrafish, a leading biomedical model for early development. Egg batches of good and poor quality, evidenced by embryo survival for 24 h, were sampled immediately after spawning and used to create pooled or replicated samp...
متن کاملDanio Rerio, the Zebrafish
The zebrafish has long been a favourite model for the study of vertebrate development. Here we provide an overview of the current state of knowledge and resources for the study of this fish, with comments on the future direction of zebrafish genomics from Professor Mark Fishman and Dr Stephen Wilson.

The herbicides used in agriculture threaten aquatic ecosystems and biodiversity on a global scale. There are several reports on the residues of currently used herbicides in the tissues of aquatic organisms. This study aimed to determine the effect of concentration on the accumulation of Butachlor in liver tissue in zebrafish fish exposed to sub-lethal concentrations of the toxin. In this study,...
متن کاملBIOLOGY OF ZEBRAFISH, Danio rerio
Zebrafish, Danio rerio (Hamilton), is one of the most important vertebrate model organism in genetics, developmental biology, neurophysiology and biomedicine (Rubinstein, 2003; Amsterdam and Hopkins, 2006). It is a small, robust fish, so large numbers can be kept easily and cheaply in the laboratory, where it breeds all year round. Development is rapid, with precursors to all major organs devel...

Paraben compounds are used widely as preservatives to inhibit microbial growth and extend used in pharmaceuticals, hygienic, cosmetics, care products. They pose the potential for reproductive toxicity for users. This paper provides an overview of the applications of histopathological biomarkers in reproductive toxicity in zebrafish caused by methylparaben. The present study investigated the cha...
